Az 5-metoxi-2-{[(4-metoxi-3,5-dimetil-2-piridil)-metil]szulfinil}-lH-benzimidazol vegyület, szabad nevén omeprazol és annak terápiásan elfogadható sói szerepelnek EP 005129 (D1) és EP 124495 (D2) sz. szabadalmainkban - lásd később. Omeprazol és alkálisói hatásosan gátolják a gyomorsav kiválasztást és gyomorfekély-gátlók, amelyeket Losec® védjegy alatt forgalmazunk.
A vegyületek (amelyek szulfinil-származékok) a kén-atomon aszimmetrikus központtal rendelkeznek és ezért az várható, hogy izomerek [R és S enantiomerek és (-) és ( ) optikai izomerek] formájában jelenhetnek meg. Nyitott kérdés azonban, hogy ezeket az enantiomereket valóban el lehet-e különíteni és hogy milyen tulajdonságokkal rendelkeznek.
Mivel ismert volt, hogy omeprazol beadása után kémiailag átalakul és egy akirális szulfénamid keletkezik (vagyis akirális vegyület) és az, hogya két enantiomer ugyanazzá a biológiailag aktív de akirális szulfénamiddá alakul - a szakember számára nem volt indokolt, hogy igen tiszta enantiomerek előállítása irányában kutasson (lásd a 3. Melléklet reakcióegyenletét.)
Ezen túlmenően az omeprazol racemizálása volt várható (vagyis az, hogy az egyik enantiomer a másikká alakul át és ez racém omeprazolt eredményez). A racemizálási felezési időt omeprazol enantiomerek esetében kb. 130 órára becsülték 37 ˚C hőmérsékleten (lásd 4. Melléklet).
Továbbá nem volt gyakorlati módszer az omeprazol enantiomerjeinek szintézisére, amellyel a szükséges tisztaságban és mennyiségben lehetett volna előállítani ahhoz, hogy gyógyszerré való fejlesztése szóbajöhessen.
A jelen találmány az omeprazol (-)-enantiomerje optikailag tiszta új alkálisóit biztosítja, mégpedig az S-(-)-omeprazol Na, K, Ca és Mg sóit stabil, szilárd alakokban, amelyek gyógyszerekben alkalmazhatók. Váratlan módon előnyös tulajdonságokkal rendelkeznek mind a racemáthoz, mind a nem-sóenantiomerekhez viszonyítva: előállíthatók szilárd állapotban ipari méretben, kiváló stabilitással rendelkeznek racemizációval szemben és előnyös biológiai tulajdonságokat mutatnak a racemátokhoz képest - még a racemát azonos alkálikus sóihoz képest is.
A találmány szerinti (-)-S-omeprazol sók igen hatásosak a nyelőcső kezelésére és jobb terápiás profilt biztosítanak azáltal, hogy csökken az eltérés a gyógyszernek az egyes betegekre gyakorolt hatása között.
Translation - English THE INVENTION
The compound 5-methoxy-2-{[4-methoxy-3,5-dimethylpyridin-2-yl)methyl]sulfinyl}-1H-benzimidazole, having the nonproprietary name omeprazole, and therapeutically acceptable salts thereof are described in our patents EP 005129 (D1) and EP 124495 (D2) (see below). Omeprazole and alkaline salts thereof, sold under our trademark LOSEC®, effectively inhibit the secretion of gastric acid and are gastric ulcer inhibitors.
The compounds (which are sulfinyl derivatives) have an asymmetric center on a sulfur atom and can therefore be expected to occur in isomeric forms [R and S enantiomers and (-) and ( ) optical isomers]. However, it is an open question whether these enantiomers can be separated indeed and what their features are.
As it was known that after its intake omeprazole chemically transforms and changes into an achiral sulfenamide (that is, an achiral compound), and that the two enantiomers transform into the same biologically active but achiral sulfenamide, there was no reason for a person skilled in the art to steer research towards producing highly pure enantiomers (see the reaction scheme in Annex 3).
Furthermore, the racemization of omeprazole was expected (i.e. that one enantimer transforms into the other, resulting in racemic omeprazole). The half time of the racemization of omeprazole enantiomers was estimated at approximately 130 hours at a temperature of 37 ˚C (see Annex 4).
Furthermore, there was no practical method for synthesizing the enantiomers of omeprazole achieving the necessary purity and quantity so that it could potentially be developed into a pharmaceutical product.
The present invention provides optically pure new alkaline salts of the (-)-enantiomer of omeprazole, namely Na, K, Ca and Mg salts of S-(-)-omeprazole in stable, solid forms, which can be used in pharmaceuticals. Unexpectedly, they have favorable features compared to both the racemate and the enantiomers of the non-salt form; they can be produced as solids on an industrial scale, have high stability against racemization, and have favorable biological features compared to racemates, even compared to the same alkaline salts of the racemate.
The (-)-S-omeprazole salts according to the invention are highly effective in the treatment of the esophagus and provide a better therapeutical profile because there are less individual differences in the effects of the pharmaceutical.
